If I decide not to put the cover on a backpack will it still be water resistant

Not sure. I’d have a cover for just in case. Depends how much moisture is occurring. Otherwise I like my backpack I use it for everyday use

Can I use this for school? Will it be able to support 20-25 lbs of books, papers, and a bunch of other school supplies with no problems?

I don’t see why not, I use it as a fishing bag, and load it to the max with no problems.
