You do not need the base game in order to play the expansion. You can play each expansion alone, but you can also combine any combinations of villains for play.

What is the deal with Ratigan's objective tile?

It changes when the Robot Queen is discarded. He has 2 different objectives.

Can this game be played by one player?

Unless you set up multiple boards and played against yourself, no. Its meant for at least 2 players. We usually play with 3-5 players.
