You could use either. But a pressure sprayer would be more economical with less waste unless your hose end sprayer is reliable in it's measurement and disbursement of the product.

That “jelly” like substance is akin to a SCOBY, like when making kombucha. Should still be good

When applying to leaf surface as a foliar, it is best wait until evening or early morning. Do not spray the diluted EM-1 directly on the fruit/vegetable or flowers. You can always spray your garden and then follow with a good watering to make sure none of the diluted EM-1 remains on the flower or fruit/vegetable if that is easier than trying to simply spray at the base of the plant.
