Q: Is the “hand wheel” a shut off valve? Is there a gas shut off valve included?

No this kit does not come with a shut off valve, it's just a regulator kit. In conjunction with this kit I bought the Mr Heater Universal Appliance Hook-up kit Accessories #F271239 which does come with a shut off valve. Depending on what you are trying to do you might need both kits or perhaps just the Accessories kit but in order to get a regulator and shut off valve I bought both kits. You can buy the regulator kit if you need the regulator and simply go to a hardware store and buy just a shut off valve. I bought the Accessories kit because it contains other useful components I thought I might need. Hope this helps.

Q: If you Were to install a shutoff valve, would you place on heater-side of hose or the propane tank end?

If put on heater side and you remove stove for summer line will stay primed.

Q: Are any adjustments needed to the 2 stage regulator for a 40 or 100lb tank?

This regulator will work with 20 30 or 100 pound tank
