Q: Is there room for a few over night clothing.

I would say yes. In the video, the diary size is A5 and the glasses case next to it fit in no bother and there is still a little room at either side. My hand fits in at the top so you still have that height above the zip. I also love the fact that the zip is at your back so it's more secure. Hope this helps.

Q: Is the material of the backpack genuine leather??

Yes, this is a genuine leather bag. Very good quality and well made

Q: Would you say it’s easy to get things in and out of the bag? If you load from the back, could you get your water bottle out without dislodging iPad ?

You can easily get things in and out, you could remove a water bottle without dislodging your iPad. The bag has a two way zip so you can choose the way you open and close the bag, you can open slightly to remove smaller items, or open completely to remove all items, you can choose to open from either side depending on what suits you.

Q: What is the difference between black and black2. ?

The leather in each bag has a different finish. I chose Black 2 because I preferred that one.
